You can turn any and all extras that you have on at any time, and they'll be effective until you turn them off. Once you unlock them, pause the game at any time and select "Extras". Once a Pink Brick is available, go to Gwenpool's Room in the Avengers Mansion and head to the closet on the left side of the room.
Whereas in the old games they used to have Red Bricks that hid in story missions, Pink Bricks are instead available in Gwenpool Missions.
There are seventeen Pink Bricks in the game: seven you have available for purchase at the outset of the game, and ten you have to find.
Extras are activated in this game via Pink Bricks.
